Supplements are not medicines
Dietary supplements are not medicines. In the United States they are regulated as food rather than drugs, which means the FDA does not review them for safety or effectiveness before they go on sale, and manufacturers cannot legally claim a supplement treats, cures, or prevents any disease. Nothing on this page is a substitute for prescription treatment, and no supplement described here has been shown to produce the weight loss seen with semaglutide or tirzepatide. If you are considering a supplement — particularly alongside prescription medication — talk to a doctor or pharmacist first, because interactions are real and some ingredients are not appropriate for everyone.
Strongest Evidence
Fermentable fiber (inulin, resistant starch, beta glucan)
The clearest mechanism in the category. Fibers reach the colon undigested, bacteria ferment them, and short-chain fatty acids stimulate GLP-1 secretion from L-cells. A study indexed at PubMed 20130660 found inulin increased postprandial serum short-chain fatty acids and reduced free fatty acids and ghrelin in healthy subjects. Published reviews conclude fermentable dietary fibers can influence GLP-1 secretion through microbiota-dependent fermentation and SCFA production.
Limitation: research doses are 5–15 g per day. Most capsules deliver a few hundred milligrams. And the GLP-1 rise is physiological and short-lived.
Psyllium
Published research (PMC10389520) describes gel-forming non-fermented psyllium, dosed just before meals, as effective in facilitating weight loss in overweight and obese participants. A separate trial found psyllium supplementation contributed to greater fullness and less hunger between meals.
Limitation: psyllium is largely non-fermented, so it does not act through the SCFA-GLP-1 route at all. It works mechanically. Effects on weight are modest.
Protein
Research indexed at PMC7539343 concluded dietary protein elevates GLP-1, CCK and PYY while decreasing ghrelin. Later work found protein stimulates GLP-1 and PYY secretion to a greater extent than carbohydrate.
Limitation: this is a dietary finding rather than a supplement one. It argues for eating more protein, not for buying a capsule.
Moderate Evidence
Berberine
The most-studied botanical here. A meta-analysis (PMC8107691) concluded berberine can improve obesity and hyperlipidemia by reducing triglycerides, total cholesterol and LDL while increasing HDL, and reduce insulin resistance. A 2025 randomized clinical trial in JAMA Network Open assessed berberine for visceral adipose tissue and liver fat in diabetes-free individuals.
Limitation: effects are metabolic and modest, not medication-scale weight loss. Requires 900–2,000 mg daily in divided doses. Significant CYP3A4 drug interactions. Documented potency variability between commercial preparations (PMC5807210).
Early or Preclinical Evidence
Akkermansia muciniphila
A specific documented mechanism: Yoon and colleagues (2021) identified the P9 protein secreted by A. muciniphila that stimulates GLP-1 secretion, and later work (PubMed 40806100) reported A. muciniphila extracts enhancing GLP-1 release, concluding they “may offer metabolic benefits.”
Limitation: largely cell and animal work. Human trials showing supplement-form A. muciniphila produces meaningful weight change do not yet exist.
Green tea catechins (EGCG), curcumin, gymnema, saffron, citrus flavonoids
Each has some published research, generally small and often supplier-funded where a branded extract is involved. Reviews of natural products boosting GLP-1 (PubMed 34981502) list berberine, tea, curcumin, cinnamon, resveratrol and others as having some supporting data.
Limitation: small trials, inconsistent results, and bioavailability problems — curcumin in particular is poorly absorbed without an enhancer. Green tea extract has been associated with rare liver injury at high doses.
How to apply this when shopping
Evidence for an ingredient transfers to a product only if the product contains that ingredient at the studied dose. Since most formulas do not publish per-ingredient amounts, the honest default assumption is that they are under-dosed.
The corollary: a cheap single-ingredient product at a published, research-relevant dose is usually a better buy than an expensive blend with an impressive ingredient list and no numbers.

Which GLP-1 supplement ingredient has the most evidence?
Berberine has the most human trial data, including meta-analyses and a 2025 randomized clinical trial in JAMA Network Open, though its evidence is metabolic rather than GLP-1-specific. For satiety, psyllium has the most direct trial support. For a GLP-1 mechanism specifically, fermentable fiber has the clearest physiological basis.
Is the evidence for these ingredients strong?
It varies enormously and it is never as strong as the evidence for prescription GLP-1 medications. The best of it — berberine meta-analyses, psyllium satiety trials, fiber and SCFA physiology — supports modest, specific effects. The weakest of it is preclinical mechanism dressed up as clinical benefit.
Does ingredient evidence apply to the product I am buying?
Only if the product delivers the ingredient at the studied dose, which most do not. This is the single most common gap in the category: a formula lists berberine, the research uses 900–2,000 mg per day, and the label does not say how much is in it. Ingredient evidence transfers to a product only when the dose does.
